Working Principle Of Ultra Micro Spectrophotometer

2025-02-08

Ultra micro spectrophotometer is a precision optical instrument that undergoes fine assembly and debugging before leaving the factory. If the instrument can be properly maintained and serviced, it can not only ensure the reliability and stability of the instrument, but also extend its service life.

Ultra micro spectrophotometers are commonly used to quantify oligonucleotides, single stranded, double stranded DNA, and RNA dissolved in buffer solutions. The absorption wavelength of the higher absorption peak of nucleic acid is 260 nm. The molecular composition of each nucleic acid is different, so its conversion factor is different. Quantify different types of nucleic acids by selecting corresponding coefficients in advance. For example, the absorbance values of 1OD are equivalent to 50 μ g/ml dsDNA, 37 μ g/ml ssDNA, 40 μ g/ml RNA, and 30 μ g/ml Olig, respectively. The absorbance value after testing is converted by the above coefficient to obtain the corresponding sample concentration.

The working principle of an ultra micro spectrophotometer is to use spectrophotometry to quantitatively and qualitatively analyze substances. It is commonly used for nucleic acid, protein quantification, and bacterial growth concentration quantification. UV visible spectrophotometers have wide and important applications in scientific research fields such as physics, chemistry, biology, medicine, materials science, environmental science, as well as in modern production and management departments such as chemical engineering, medicine, environmental testing, metallurgy, etc.
